In elderly, skin becomes dryness, rougher, wrinkling, sagging and incremental laxity of the skin caused by increased activation of extracellular matrix disruption enzymes, oxidative stress and DNA damage. Searching of ingredients from natural sources in order to restrain these mechanisms, have been attending because of their competitive effectiveness and lower toxicity effects. Among of natural active ingredients, mushroom provide a wide spectrum of biological activity and could be included into the dietary supplements, pharmaceutical and especially, cosmeceutical potential. The present study has been attempted to extract and evaluate the biological activity of various edible mushroom such as Coprinopsis cinerea, Cordyceps militaris, Ganoderma lucidum etc. Each mushroom was extracted by ethanol (95%) and water. The total phenolic, anti-oxidant activity and anti-tyrosinase inhibitory activity were investigated using Folin-Ciocalteu method, DPPH radical scavenging assay and Dopachrome method, respectively. Among of these mushrooms, the alcoholic extracts of Coprinopsis cinerea were found to be more activities. IC50 values observed in DPPH and anti-elastase activity were 0.56±0.21 mg/ml and 0.56±0.21 mg/m, respectively. The anti-tyrosinase and anti-melanogenesis activity revealed to be 79.84 ± 9.71 % at 10mg/ml and 122.45±4.79 % at 1mg/ml respectively. Hence, this extract was incorporated into liposome to enhance its skin penetration ability. The liposomal formulations composed of phospholipid, cholesterol and Coprinopsis cinerea extract were prepared using a thin film hydration method. The entrapment efficiency of obtained Coprinopsis cinereal extract loaded liposome was 97.6%. The vesicle size and zeta-potential were found to be 80-200 nm and -12.8 ± 0.8 mV, respectively. Significantly higher skin permeation of liposome has been achieved, as compared to solution. Results of cytotoxicity indicated both of extract and extract loaded- liposome were safe. Then, extract loaded- liposome was incorporated into cosmeceutical products i.e., anti-aging nanocream and whitening nanoserum. Under the study conditions on normal skin without spot, after 28 days of twice-daily use, the whitening nanoserum increase in skin clarity 95% of the subjects and decrease in skin pigmentation 59% of the subjects. For anti-aging nanocream, anti-wrinkle and smoothing effect significant decrease in the average roughness (Ra) and relief amplitude (Rt) but increase in the cutaneous hydration rate. Finally, acute cutaneous tolerance was investigated by single patch test, indicating that both products are non-irritating. The results suggest that Coprinopsis cinerea extract loaded-liposome can be considered as new natural sources to be potentially used as whitening agent or anti-aging ingredients in skin care formulations. Summary: ในงานวิจัยนี้ได้คัดเลือกเห็ดโดยสกัดและศึกษาฤทธิ์ทางชีวภาพของเห็ดที่รับประทานได้ชนิดต่างๆ เช่น เห็ดโคนน้อย เห็ดถังเช่า เห็ดหลินจือ เป็นต้น โดยนำมาสกัดด้วยน้ำและแอลกอฮอร์ร้อยละ 95 แล้วนำมาศึกษาฤทธิ์ต้านปฏิกิริยาออกซิเดชัน ฤทธิ์ต้านการทำงานของเอนไซม์ไทโรซิเนส โดยใช้วิธีฟอลินซิโอแคลตู วิธีต้านอนุมูลอิสระดีพีพีเอช และวิธีโดพาโครม ตามลำดับ นอกจากนี้ได้ศึกษาฤทธิ์ยับยั้งเอนไซม์อิลาสเทส ฤทธิ์ยับยั้งการเจริญของเซลล์เมลาโนไซต์ และความเป็นพิษต่อเซลล์ผิวหนังมนุษย์ โดยวิธีการเพาะเลี้ยงเซลล์ในหลอดทดลอง ซึ่งสารสกัดแอลกอฮอล์ของเห็ดโคนน้อยมีฤทธิ์ทางชีวภาพที่โดดเด่นกว่าเห็ดชนิดอื่นที่ทำการทดสอบโดยสารสกัดแอลกอฮอลล์ของเห็ดโคนน้อย สามารถยับยั้งสารอนุมูลอิสระได้คิดเป็นร้อยละ 50 (IC50) ที่ความเข้มข้น 0.56±0.21 มิลลิกรัม/มิลลิลิตร และมีฤทธิ์ยับยั้งเอนไซม์อิลาสเทสได้ ร้อยละ 50 ของตัวอย่างทดสอบ (IC50) ที่ความเข้มข้น 0.029±0.003 มิลลิกรัม/มิลลิลิตร ซึ่งให้ผลดีกว่าสารมาตรฐานอิพิแกลโลคาเทชินแกลเลท (epigallocatechin gallate; EGCG) 7 เท่า มีฤทธิ์ยับยั้งเอนไซม์ไทโรซิเนสได้ร้อยละ 79.84 ± 9.71 ที่ความเข้มข้น 10 มิลลิกรัม/มิลลิลิตร และยับยั้งการเจริญของเซลล์ที่สร้างเม็ดสี หรือเซลล์เมลาโนไซต์ร้อยละ 122.45±4.79 ที่ความเข้มข้น 1 มิลลิกรัม/มิลลิลิตร ไม่ก่อให้เกิดความเป็นพิษต่อเซลล์ผิวหนังมนุษย์ เมื่อนำสารสกัดแอลกอฮอลล์ของเห็ดโคนน้อยมากักเก็บในรูปแบบลิโพโซมมีขนาดอนุภาคน้อยกว่า 100 นาโนเมตร สามารถช่วยเพิ่มความคงสภาพของสารสกัดและประสิทธิภาพการซึมผ่านผิวหนังได้ 8 เท่า เมื่อเปรียบเทียบกับสารสกัดที่ไม่ได้กักเก็บ และสามารถนำลิโพโซมที่บรรจุสารจากเห็ดโคนน้อยมาพัฒนาเป็นผลิตภัณฑ์เวชสำอางได้สองผลิตภัณฑ์คือผลิตภัณฑ์นาโนซีรัมบำรุงผิวหน้า และผลิตผลิตภัณฑ์นาโนครีมเพื่อเพิ่มความชุ่มชื้นและลดเลือนริ้วรอย โดยผลิตภัณฑ์นาโนซีรัมบำรุงผิวหน้า เพื่อผิวกระจ่างใส ลดเลือนจุดด่างดำ ที่พัฒนาขึ้น มีประสิทธิภาพในการลดเลือนจุดด่างดำให้มีจำนวนลดลงร้อยละ 85 มีขนาดเล็กลงร้อยละ 91 และทำให้ผิวกระจ่างใสขึ้นร้อยละ 97 ภายในระยะเวลา 28 วันของการทดสอบ และผลิตผลิตภัณฑ์นาโนครีมเพื่อเพิ่มความชุ่มชื้นและลดเลือนริ้วรอยมีความเข้มข้นของพอลิแซ็กคาไรด์ของเห็ดโคนน้อย ร้อยละ 0.1-5 (น้ำหนัก/น้ำหนัก) และสารสกัด แอลกอฮอลล์ของเห็ดโคนน้อย ร้อยละ 0.1-5 (น้ำหนัก/น้ำหนัก) มีประสิทธิภาพทำให้ผิวเรียนเนียนขึ้นโดยลดความขรุขระของผิวได้ ร้อยละ 8 สามารถลดเลือนริ้วรอยบนผิวเฉลี่ยได้ ร้อยละ 7 และสามารถเพิ่มความชุ่มชื้นให้ผิวได้ ร้อยละ 15 เมื่อเปรียบเทียบกันครีมพื้นภายในระยะเวลา 28 วันของการทดสอบ และผลิตภัณฑ์ทั้งสองไม่ก่อให้เกิดการระคายเคืองต่อผิวหนังในอาสาสมัคร จากผลการศึกษาแสดงให้เห็นว่า สารสกัดเห็ดโคนน้อยที่กักเก็บในลิโพโซม เป็นแหล่งสารสำคัญจากธรรมชาติแหล่งใหม่ที่มีศักยภาพสามารถนำมาใช้เป็นสารเพื่อความขาวและต้านริ้วรอยในผลิตภัณฑ์เครื่องสำอางได้
