Based on the information from various brands of jelly products in the market, it was found that soft jelly, chewy texture, is more suitable than gummy jelly due to the short time and low temperature for production used as a prototype of food products fortified of probiotics or food ingredients. Regarding the soft jell production process, there are important factors related to the characteristics of low-sweet health jelly products such as pH value, types and amount of gelling agents, and the amount of sugar substitute (maltitol syrup). The results showed that the increased pH value affects the significant difference of soft jelly texture in terms of hardness and cohesiveness increasing (p<0.05). The increased amount of hydrocolloids (carrageenan and konjac powder) affects the significant difference of texture in terms of hardness, springiness, and gumminess increasing (p<0.05). The using of maltitol syrup produces a softer jelly than that of sucrose. Moreover, the addition of maltitol syrup tended to increase the total soluble solids of soft jelly (p<0.05), but no effect changing in soft jelly texture was found. The maltitol syrup can be used up to 15%. Thereafter, the prototype was developed in the form of a soft jelly sheet fortified with probiotics (Lactobacillus paracasei. KT5 TISTR 2688) at 1.0 %, 1.5% and 1.75% with 5% yogurt powder The physicochemical properties, the amount of probiotics, and sensory evaluation will be evaluated The results showed that soft jell sheet fortified with 1.75 % probiotic and added flavor with 5% yogurt powder had probiotic content of 1.57 x 107 according as specified by the Food and Drug Administration (FDA Thailand) (1x106 CFU/g) and received the overall liking scores in the range of 6.05 ±0.60 corresponding to “like moderately” with 100% acceptability. After that, the selected sample was analyzed for nutritional value and shelf life study. The 4-week shelf life study of soft jelly sheet results showed that the amount of probiotic bacteria in the product with preservative (potassium sorbate 500 ppm) ranged 1.30 x107-1.31x107 CFU/g while the jelly product without preservative have the probiotic content from 8.9x106-1.80 x107 CFU/g which are in the standard of FDA Thailand. Regarding sensory evaluation, the product still obtains 90-100% acceptability and a score of overall liking in the level of “like slightly to like moderately” (5.22±0.86 scores) The nutritional value per serving of product (2 pieces/20 grams) provides 15 kcal of energy without sugar and cholesterol. The production cost of jelly sheet products fortified with probiotic about 38.38 Baht/pack (60 g/pack). The cup jelly fortified with probiotics (Bifidobacterium pseudolongum M2 TISTR 2237) has been developed. The study of commercially jelly products and sensorial evaluation by trained panelists showed that jelly with soft, clear and springy texture is more acceptable than that of firm and brittle texture. Referring the flavor of most commercial jellies, pineapple flavor has not been found Therefore, this information is used in developing the prototype product of cup jelly enhanced with probiotics which is in the developing process of product enriched with probiotics by encapsulation technique of probiotic cells into the bead form which will be added in the cup jelly at 10%. The results of shelf life study of pineapple jelly fortified with probiotic beads at 4 C for 4 weeks showed that the amount of probiotic bacteria in the product with preservatives (the ration of potassium sorbate and sodium sorbate at 1:1) 500 ppm was 7.5 x107 CFU/g while the product without preservative had the probiotic bacteria content had shelf life of 3 weeks (8.9x106 CFU/g) which are in the standard of FDA. The product receives the scores of overall liking from the panelists
in the level of “like slightly to like moderately” (5.89±0.76 scores) and still obtains 100% acceptability by sensory evaluation. The nutritional value per serving of product (1 cup/100 g) had 70 kcal, 21.86 g of calcium and no cholesterol. The pineapple jelly had the production cost about 41.89 Baht/ cup (100 g) without packaging and labor costสาระสังเขป: จากการศึกษาข้อมูลผลิตภัณฑ์เยลลี่ในท้องตลาด สามารถแบ่งได้ 2 รูป ได้แก่ เยลลี่แห้ง และ เยลลี่ชนิดนุ่ม โดยได้ข้อมูลจากการเลือกซื้อผลิตภัณฑ์เยลลี่ที่หลากหลายยี่ห้อในท้องตลาดทำให้พบว่าเยลลี่ที่มีเนื้อสัมผัสนุ่ม สู้ฟัน มีความเหมาะสมมากกว่าเยลลี่แห้ง เนื่องจากใช้เวลาสั้นและความร้อนที่มีอุณหภูมิต่ำในการผลิต เหมาะกับการเป็นต้นแบบผลิตภัณฑ์อาหารเสริมโพรไบโอติก เพื่อสุขภาพพร้อมบริโภค หรือสำหรับใช้เป็นส่วนผสมของอาหาร ทั้งนี้ในกระบวนการผลิตผลิตภัณฑ์เยลลี่เบื้องต้นมีปัจจัยสำคัญที่เกี่ยวข้องกับคุณลักษณะของผลิตภัณฑ์เยลลี่ ได้แก่ ค่าความเป็นกรด-เบส (pH) ชนิดและปริมาณสารประกอบไฮโดรคอลลอยด์ และปริมาณสารให้ความหวานทดแทนน้ำตาล (มอลทิทอลไซรัป) จากการทดลองพบว่า ค่า ความเป็นกรด-เบสที่เพิ่มขึ้น มีผลต่อความแตกต่างอย่างมีนัยสำคัญทางสถิติของเนื้อสัมผัสเยลลี่ชนิดนุ่มในด้านค่าความแข็ง (Hardness) และค่าความยึดเกาะ (Cohesiveness) ที่เพิ่มขึ้น (p<0.05) ส่วนการเพิ่มขึ้นของปริมาณสารประกอบไฮโดรคอลลอยด์ (กลุ่มคาราจีแนนและผงบุก) มีผลต่อความแตกต่างอย่างมีนัยสำคัญทางสถิติของเนื้อสัมผัส ในด้านค่าความแข็งค่าความยืดหยุ่น (Springiness) และค่าพลังงานที่ทำให้ชิ้นเยลลี่แตกออก (Gumminess) ของเยลลี่เพิ่มขึ้น (p<0.05) การใช้มอลทิทอลไซรัปทำให้ได้เจลที่มีเนื้อนุ่มกว่าการใช้น้ำตาลซูโครส นอกจากนี้ การเพิ่มปริมาณมอลทิทอลไซรัป มีแนวโน้มส่งผลให้ปริมาณของแข็งที่ละลายน้ำทั้งหมดเพิ่มขึ้น (p<0.05) แต่ไม่มีผลต่อการเปลี่ยนแปลงลักษณะเนื้อสัมผัสของเยลลี่ชนิดนุ่ม ทั้งนี้สามารถใช้มอลทิทอลไซรัปได้ถึงร้อยละ 15 เมื่อได้ผลิตภัณฑ์ต้นแบบแล้ว จึงนำมาพัฒนาผลิตภัณฑ์อาหารรูปแบบของเยลลี่นุ่มแบบแผ่น โดยการเติมเชื้อโพรไบโอติก (Lactobacillus paracasei KT5 TISTR 2688) ในปริมาณร้อยละ 1.0, 1.5 และ 1.75 และศึกษาคุณภาพทางด้านกายภาพ เคมี ปริมาณเชื้อโพรไบโอติก การประเมินผลทางประสาทสัมผัส พบว่า เยลลี่แบบแผ่น เสริมโพรไบโอติกร้อยละ 1.75 และแต่งกลิ่นรสโดยเติมผงโยเกิร์ต ร้อยละ 5 มีปริมาณเชื้อโพรไบโอติก 1.57 x 107 ตามที่สำนักงานคณะกรรมการอาหารและยาของประเทศไทย (อย.) กำหนด (1x106 โคโลนี/กรัม) และได้รับคะแนนความชอบโดยรวม 6.05 ±0.60 ในระดับความชอบปานกลาง และการยอมรับร้อยละ 100 และจากนั้นศึกษาอายุการเก็บรักษา และวิเคราะห์คุณค่าทางโภชนาการของผลิตภัณฑ์ ผลการศึกษาอายุการเก็บรักษาของเยลลี่แบบแผ่น 4 สัปดาห์ ที่ 4 องศาเซลเซียส พบว่าปริมาณเชื้อโพรไบโอติกของผลิตภัณฑ์ที่ใส่วัตถุกันเสีย (โพแทสเซียมซอร์เบท 500 ppm) มีปริมาณเหลือรอดอยู่ในช่วง 6.5x106- 1.30 x107 โคโลนี/กรัม ขณะที่ผลิตภัณฑ์เยลลี่แผ่นเสริมโพรไบโอติกที่ไม่ใส่สารกันเสียมีปริมาณเหลือรอดอยู่ในช่วง 6.0x106-1.80x107 โคโลนี/กรัม ซึ่งยังอยู่ในช่วงที่สำนักงานคณะกรรมการอาหารและยาของประเทศไทย (อย.) กำหนด (1x106 โคโลนี/กรัม) ผลิตภัณฑ์ยังคงได้รับการยอมรับทางประสาทสัมผัส ร้อยละ 90-100 และได้รับคะแนนความชอบโดยรวม ในระดับ “ความชอบเล็กน้อย-ปาน-กลาง” (คะแนน 5.22±0.86) ส่วนคุณค่าทางโภชนาการต่อ หน่วยบริโภค (2 ชิ้น/ 20 กรัม) ให้พลังงาน 15 กิโลแคลอรี่ ไม่มีน้ำตาล ไม่มีคอเลสเตอรอล ต้นทุนการผลิตของผลิตภัณฑ์เยลลี่แผ่นเสริมโพรไบโอติก อยู่ที่ 38.38 บาท/ซอง (ซองละ 60 กรัม) ได้ดำเนินการพัฒนาผลิตภัณฑ์เยลลี่เสริมโพร-ไบโอติก Bifidobacterium pseudolongum M2 TISTR 2237 แบบเนื้อสัมผัสนุ่มพร้อมบริโภค จากการศึกษาข้อมูลผลิตภัณฑ์ทางการค้า และทดสอบชิมจากผู้ทดสอบที่ได้รับการฝึกฝน พบว่า เยลลี่ที่ให้เนื้อสัมผัสนุ่ม ใส มีความยืดหยุ่น ได้รับการยอมรับมากกว่า เนื้อแน่น เปราะ ส่วนรสชาติของเยลลี่ทางการค้าส่วนใหญ่ ยังไม่พบรสสับปะรด ดังนั้นจึงได้นำข้อมูลเหล่านี้มาใช้ในการพัฒนาผลิตภัณฑ์ต้นแบบเยลลี่บรรจุถ้วยเสริมโพรไบโอติก โดยทำการห่อหุ้มเซลล์ของเชื้อโพรไบโอติก (encapsulation) ให้อยู่ในรูปเม็ดบีดส์ แล้วเติมในผลิตภัณฑ์เยลลี่ ในปริมาณร้อยละ 10 จากผลการศึกษาอายุการเก็บรักษาของเยลลี่รสสับปะรดแบบบรรจุถ้วยเสริมเม็ดบีดส์โพรไบโอติก ที่ 4 องศาเซลเซียส สามารถเก็บได้ 4 สัปดาห์ พบว่าปริมาณเชื้อโพรไบโอติกของผลิตภัณฑ์ ที่ใส่วัตถุกันเสีย (อัตราส่วน โพแทสเซียมซอร์เบทและโซเดียมเบนโซเอท ที่ 1 : 1) 500 ppm เหลือรอด 7.5 x107 โคโลนีต่อกรัม ขณะที่ผลิตภัณฑ์ ไม่ใส่วัตถุกันเสีย สามารถเก็บได้ 2 สัปดาห์ (8.9 x106 โคโลนี/กรัม) ซึ่งยังอยู่ในช่วงมาตรฐานที่ อย. กำหนด จากผลการทดสอบทางประสาทสัมผัส ผลิตภัณฑ์ได้รับคะแนนความชอบโดยรวมจากผู้ทดสอบ ในระดับ “ความชอบเล็กน้อย-ปานกลาง” (คะแนน 5.89±0.76) และยังคงได้รับการยอมรับ ร้อยละ 100 ส่วนคุณค่าทางโภชนาการต่อหน่วยบริโภค (1 ถ้วย/ 100 กรัม) ให้พลังงาน 70 กิโลแคลอรี่ มีปริมาณแคลเซียม 21.86 กรัม และไม่มีคอเลสเตอรอล ต้นทุนการผลิตของผลิตภัณฑ์อยู่ที่ 41.89 บาท/ ถ้วย (100 กรัม) ซึ่งไม่รวมค่าบรรจุภัณฑ์ และค่าแรง
