In this study, gene expression analysis of drought stress related genes in J. curcas including PIP (plasma membrane intrinsic protein), NAC transcription factor (NAM, ATAF, and CUC), DREB1A (dehydration responsive element binding protein 1A) and ZEP (zeaxanthin epoxidase) genes were analyzed by quantitative real-time PCR. Strong induction of DREB1A gene was observed under drought stress conditions. The DREB1A gene was then further cloned and sequenced. The DREB1A clone was isolated from drought stressed J. curcas using reverse transcriptase polymerase chain reaction (RT-PCR) and 5'-3' rapid amplification of cDNA end (5'-3' RACE). The full length of DREB1A contained 678 bp open reading frame (ORF), translated for 225 amino acids. The nucleotide and amino acid sequences showed 99% sequence identity with the J. curcas DREB1A gene (accession number XM012234936.1) and the J. curcas DREB1A protein (accession number XP012090326.1), respectively. The NcoI and SpeI restriction sites were then added to the 5' and 3' ends of the DREB1 gene by PCR technique. The DREB1A fragment was cloned into the pCAMBIA1305.1 between CaMV35S promoter and NOS terminator in the positions of NcoI and SpeI restriction sites. Protocols for callus initiation, plantlet regeneration, and transformation were also developed.
