In this project, several extraction methods of dried C. indicum L. flowers were studied such as steam distillation, water extraction, solvent extraction and supercritical fluid extraction (SFE) with CO2. Chemical compositions of these extracts were analyzed by HPLC method. Luteolin-7-glucoside was found as the major compound in most of these extracts except in essential oil extract and used as a marker for quality control both in the extract and finishing product. Pharmacological studies of these extracts were investigated for further applications. The water extract of C. indicum showed a significant (p<0.05) increasing in sleeping duration and muscle relaxant activity. This extract was developed to a sedative food supplement product. The safety evaluation of the extract and the product were also investigated. Acute oral toxicity showed that LD50 of the extract was above 2,000 mg/kg body weight while LD50 of the product was above 15,000 mg/kg body weight. These results suggested that C. indicum extract should be used as an active ingredient in sedative food supplement.สาระสังเขป: เก๊กฮวย Chrysanthemum indicum L. มีดอกสีเหลืองนิยมใช้ชงเป็นชาและเครื่องดื่มโครงการนี้ได้ศึกษาการสกัดดอกเก๊กฮวยแห้งหลายวิธี ได้แก่ การกลั่นด้วยไอน้ำ, การสกัดด้วยน้ำ, การสกัดด้วยตัวทำละลายและการสกัดด้วยคาร์บอนไดออกไซด์ในสภาวะวิกฤตยิ่งยวด และศึกษาองค์ประกอบทางเคมี โดยใช้วิธี HPLC พบว่า สารสกัดเกือบทุกชนิดยกเว้นน้ำมันหอมระเหยเก๊กฮวย มีสาร Luteolin-7-glucoside เป็นองค์ประกอบหลัก จึงใช้สารนี้ในการควบคุมคุณภาพสารสกัด และผลิตภัณฑ์ การศึกษาทางเภสัชวิทยาศาสตร์เพื่อหาประสิทธิภาพของสารสกัดเก๊กฮวยชนิดต่างๆ พบว่าสารสกัดเก๊กฮวยด้วยน้ำสามารถเพิ่มช่วงการนอนหลับและผ่อนคลายกล้ามเนื้อได้อย่าง มีนัยสำคัญทางสถิติ (p<0.05) จึงได้นำสารสกัดนี้ไปพัฒนาเป็นผลิตภัณฑ์เสริมอาหารเพื่อคลายเครียด การประเมินความปลอดภัยของสารสกัดและผลิตภัณฑ์ได้ทำการทดสอบความเป็นพิษเฉียบพลันทางปาก พบว่า สารสกัดเก๊กฮวยมีค่า LD50 มากกว่า 2,000 มิลลิกรัมต่อกิโลกรัมน้ำหนักตัว ส่วนผลิตภัณฑ์มีค่า LD50 มากกว่า 15,000 มิลลิกรัมต่อกิโลกรัมน้ำหนักตัว นับว่าสารสกัดและผลิตภัณฑ์มีประสิทธิภาพและความปลอดภัยเหมาะสำหรับใช้เป็นผลิตภัณฑ์เสริมอาหารเพื่อคลายเครียด.
