Spilanthes acmella (L.) Murr. (para cress; SA), containing limonene, was extracted and used to study pharmacological effects. The results showed that 3% w/w SA significantly possessed analgesic and anesthetic activities in rats as well as EMLA, a topically local anesthetic drug. Moreover, anti-inflammatory effect in rat ear edema model was significantly found in 3% w/w SA as well as phenylbutazone. Then, the evaluation of transdermal delivery of 3% w/w SA was manipulated to determine the maximal levels of limonene in blood circulation. The study revealed that using 3% w/w SA together with an ultrasonic device was found the highest levels of limonene in rabbit's blood at 60 minutes after topical administration. On the other hand, limonene reached maximal levels in blood at 120 minutes after using 3% w/w SA without an ultrasonic device. This finding suggests that using 3% w/w SA together with an ultrasonic device delivers the compound into the circulating system faster. Hence, SA extract was formulated in gel form, and was named as "Ac-Anes". Without irritation and sensitization in animal models, this gel was safe for skin. For quality control of the product, there was no contamination of pathogens and heavy metals in "Ac-Anes". Satisfaction survey on using "Ac-Anes" with/without an ultrasonic device in volunteers was also studied. Using "Ac-Anes" without an ultrasonic device exhibited in acceptable - satisfied level, while using "Ac-Anes" with an ultrasonic device showed in satisfied - very satisfied level. The purpose of this study aims to do the research and development on a natural product for local anesthetic activity from plant which is finally used together with an ultrasonic device in order to be an alternative drug สาระสังเขป: การวิจัยในครั้งนี้มีวัตถุประสงค์เพื่อวิจัยและพัฒนาผลิตภัณฑ์จากสมุนไพรที่มีฤทธิ์เป็นยาชาเฉพาะที่เพื่อการใช้ร่วมกับอุปกรณ์นำส่งยาชนิดที่เป็น ultrasonicอัลตราโซนิก ให้เป็นอีกทางเลือกหนึ่งสำหรับผู้ที่จะต้องทำการผ่าตัด เช่น การผ่าตัดเล็ก การทำศัลยกรรมเสริมความงาม การทำเลเซอร์เฉพาะจุดบนใบหน้า โดยนำผักคราดหัวแหวนที่มีสารสำคัญ คือ limonene มาสกัด แล้วนำมาศึกษาฤทธิ์ทางเภสัชวิทยาในหนูขาว พบว่า สารสกัดผักคราดหัวแหวน ขนาด 3% w/w สามารถบรรเทาอาการเจ็บปวด (analgesic activity) และระงับความรู้สึก (anesthetic activity) ได้อย่างมีนัยสำคัญทางสถิติ เช่นเดียวกับยามาตรฐาน EMLA. นอกจากนี้ยังพบว่ามีฤทธิ์ในการต้านการอักเสบที่ใบหูหนูขาวได้อย่างมีนัยสำคัญเช่นเดียวกับสารมาตรฐาน phenylbutazone เมื่อศึกษาการซึมผ่านผิวหนังของสารสกัดผักคราดหัวแหวนขนาด 3% w/w โดยการวัดระดับ limonene ในเลือด พบว่า เมื่อใช้ร่วมกับเครื่องอัลตราโซนิก ultrasonic สามารถตรวจพบ limonene สูงสุดในเลือดกระต่ายที่ 60 นาที ในขณะที่การทาแบบปกติอยู่ที่ 120 นาที ซึ่งแสดงให้เห็นว่าการใช้ร่วมกับเครื่องนำส่งยาชนิดอัลตราโซนิก ultrasonic สามารถผลักดันยาให้เข้าสู่กระแสเลือดได้เร็วกว่าการทาแบบปกติถึง 1 เท่าตัว จึงได้นำมาพัฒนาเป็นผลิตภัณฑ์เจลใสทาภายนอก ให้ชื่อผลิตภัณฑ์ว่า "Ac-Anes" ที่ผ่านการประเมินความปลอดภัยต่อผิวหนังสัตว์ทดลอง โดยพบว่า ไม่ก่อให้เกิดความระคายเคืองและการแพ้ใดๆ การตรวจการปนเปื้อนโลหะหนักและเชื้อจุลินทรีย์ไม่เกินค่ามาตรฐาน การศึกษาความพึงพอใจของการใช้ผลิตภัณฑ์แบบทาแล้วปิดไว้อยู่ที่ปานกลางถึงมาก ส่วนการใช้ร่วมกับเครื่องนำส่งยาชนิดอัลตราโซนิก ultrasonic มีระดับความ พึงพอใจอยู่ที่มากถึงมากที่สุด.
